***This test requires approval by the Lab Medicine resident on-call (206) 598-6190.*** For evaluating patients for diseases of immediate hypersensitivity, the preferred screening test is serum tryptase (see Lab mnemonic RTRYPT). Test results highly dependent on proper sample collection. Histamine may be released ex vivo by passing basophils in blood through a small-bore needle under vacuum or when blood clots. Serum should not be used in order to avoid the artifactual release of histamine from basophils that can occur during clotting.

One 3 mL lavender (EDTA) or pink (K2EDTA). Specimens must be collected in a pre-chilled tube and on ice.

Processing
- Processing
Centrifuge at 2-8°C and separate upper two-thirds of plasma within 20 minutes. Transfer 1 mL plasma to a plastic tube. Freeze immediately at -20°C.
