Blood (EDTA Plasma)
The UW Clinical Virology Laboratory in the Department of Laboratory Medicine and Pathology incorporates the FDA approved Roche cobas® BKV Quantitative assay for the detection of BK polyomavirus (BKV) using real-time PCR amplification.
The lower limit of detection, LoD (the minimum virus level that gives a positive result in 95% of replicates), and the lower limit of quantitation, LLoQ, are both 21.5 IU/mL.
A conversion factor of 1.1 Log10 IU/mL is equivalent to 1.0 Log10 copies/mL.
The Roche cobas® BKV assay uses primers specific for the BKV VP2 region and the BKV small t-antigen region.

Blood: 3 mL EDTA plasma or PPT
Unacceptable samples: Plasma collected in sodium or lithium heparin.
Note: Separate specimens MUST be submitted when both Serology and PCR testing are ordered.
Freshly drawn whole blood specimens may be held at 2 to 25°C for up to 24 hours prior to plasma preparation through centrifugation.
Blood: Centrifuge upon arrival, remove plasma from cells.
Plasma specimens may be stored and/or transported:
Outside Laboratory: Ship samples on dry ice. If freezer and/or dry ice is unavailable, ship immediately at 2-8°C (ice pack). Shipment time must not exceed 48 hours from collection. Sample may be stored at 2-8°C (ice pack) for up to 6 days of collection. Do not freeze/thaw.
