The detection of HSV1 and HSV2 IgG class antibodies by Western blot in serum or CSF. HSV1 and HSV2 proteins from detergent lysates ("Bernstein's lysate") of HSV infected cells are separated by electrophoresis and transferred to nitrocellulose paper. The strips of paper or "blots" containing separated fixed proteins from either HSV-1 or HSV-2 are incubated with the patient's serum. Antibodies, which bind to the viral proteins, are detected by an enzyme-mediated color change. The pattern of staining on the two blots (HSV-1 and HSV-2) is dictated by the number and identity of the HSV proteins to which the patient's immune system has antibody.
This test is reported as:
- HSV1 - Positive, Negative or Indeterminate
- HSV2 - Positive, Negative or Indeterminate
This test was developed and its performance characteristics determined by the University of Washington Department of Laboratory Medicine and Pathology. It has not been cleared or approved by the US Food and Drug Administration.

Ordering & Collection
- Specimen Type
- Blood or CSF (see instruction under test description for ordering CSF testing).
Blood: 5 mL blood in GOLD TOP serum separator tube (SST).
Also acceptable: 5 mL blood in ORANGE TOP rapid serum tube (RST) or RED TOP tube
CSF: 1 mL in sterile container (CSF antibody testing will be performed ONLY when paired with a serum sample).

Blood: Harvest sera from whole blood within 24 hours of collection. Sera may be stored at 2-8°C (ice pack) for up to 7 days. If testing will be delayed more than 7 days from collection freeze at -10°C or colder. Do not freeze/thaw.
CSF: If CSF has cellular material, centrifuge & aliquot clear CSF. CSF may be stored at 2-8°C (ice pack) for up to 7 days. If testing will be delayed more than 7 days from collection freeze at -10°C or colder. Do not freeze/thaw. Do not freeze/thaw.
Freeze samples at -10°C or colder. Ship on dry ice, do not freeze/thaw. If freezer and/or dry ice is not available, ship immediately at 2-8°C (ice pack), shipment time must not exceed 7 days from collection. Samples stored at 2-8°C greater than 7 days old will be rejected.
Unacceptable samples: Cadaver specimens or body fluids other than human sera or CSF.
