Description

Useful For:

  • Assessment of adults with cognitive impairment being evaluated for Alzheimer disease (AD) and other causes of cognitive impairment.
  • Measurement of beta-amyloid (1-42)(Abeta42), total-Tau, and phosphorylated-Tau (p-Tau) in cerebrospinal fluid (CSF) is useful in the differential diagnosis of Alzheimer Disease (AD) and other causes of cognitive impairment.

Patient Preparation: For 12 hours before specimen collection, patient should avoid taking multivitamins or dietary supplements containing biotin (vitamin B7), which is commonly found in hair, skin, and nail supplements and multivitamins.

Note: Samples for this test must be collected using the tubes provided in the Alzheimer's Disease Evaluation Collection Kit (Mayo Supply #T836). Refer to collection instructions.

Patient Preparation: For 12 hours before specimen collection, patient should avoid taking multivitamins or dietary supplements containing biotin (vitamin B7), which is commonly found in hair, skin, and nail supplements and multivitamins.

Collection Instructions:

  • Obtain an Alzheimer's Disease Evaluation CSF Collection Kit (Mayo T836) from UW-MT or HMC Specimen Processing Services.
  • Perform lumbar puncture and discard the first 1 to 2 mL of cerebrospinal fluid (CSF).
  • Collect 2.5 mL CSF directly into the Sarstedt CSF False Bottom Tube (63.614.625)*. Do not aliquot.
    • *Blue Top (CSF AD Biomarker) Sarstedt tubes from previously distributed Mayo kits will be accepted until all remaining tubes are used.
    • Also Accepted: Sarstedt 72.703.600 (1.5 mL) or Sarstedt 72.694.600 (2 mL) tubes
  • Collect 1 mL CSF directly into the Sarstedt 74.694.600 screw-cap polypropylene tube (SMART Tube name: CSF PPV).
  • If unable to obtain sufficient volume of CSF for both tubes, prioritize collection in the Mayo Sarstedt CSF False Bottom Tube. 1.5 mL is required by Mayo for testing.
  • Avoid using polystyrene collection tubes prior to transfer into approved collection tube types. Failure to adhere to this collection instruction may result in falsely low Abeta42 concentrations.
Approval Required
Orders from the Harborview Neurology Clinic (NEUROH) and the UW Multiple Sclerosis Clinic (MSCNW) are pre-approved. Orders from all other locations require LMR approval.
Handling Instructions

Outside Laboratories:

  • Obtain an Alzheimer's Disease Evaluation CSF Collection Kit (Mayo T836) by contacting Client Support Services at 206.520.4600.
  • Perform lumbar puncture and discard the first 1 to 2 mL of cerebrospinal fluid (CSF).
  • Collect 2.5 mL CSF directly into the Sarstedt CSF False Bottom Tube (63.614.625)*. Do not aliquot.
  • Refrigerate sample while awaiting shipment and transport with a cold pack.

Stability: Refrigerated (preferred): 14 days; Frozen: 60 days; Ambient: 12 hours.

Reject Due To: Gross hemolysis, gross icterus, wrong collection tube.

Receiving Instructions

If only one sample is received, prioritize the Sendout battery RCADEB. Do not aliquot CSF for the UW214.

  • RCADEB (Sarstedt CSF False Bottom or CSF AD Biomarker collection tube): Refrigerated UW-MT Mayo Sendouts rack.
  • UW214 (Sarstedt 74.694.600 screw-cap polypropylene tube, "CSF PPV"): UW-MT Chemistry -70°C Rack (UCGEN/SP58)
    • Note: If samples are submitted in any tube type besides the Mayo Sarstedt CSF False Bottom Tube (63.614.625), Mayo Blue top (CSF AD Biomarker), Sarstedt 72.703.600 (1.5 mL), or Sarstedt 72.694.600 (2 mL) tubes, contact the Lab Medicine Resident on-call for further instruction.

Note: Fisherbrand 12x75 polypropylene Cat No. 14-956-1C will be accepted if the specimen volume fills at least 80% of the tube (4 mL).
