Broad range PCR and Sanger sequencing can be used to perform accurate genus- or species-level taxonomic assignment for several fungal gene targets. Unfortunately, when PCR amplification is performed on polymicrobial specimens, the presence of multiple templates causes interference during Sanger sequencing such that results are generally uninterpretable. In contrast, next-generation sequencing methods provide individual sequence data for large numbers of DNA templates, allowing each to be classified independently. This permits fungal species identification for samples from which usable data cannot be obtained by conventional clinical methods.
